This helped us understand how HARs might have developed in the first place, according to research main author Katie Pollard.

Pollard had discovered, about two decades ago, the regions of DNA known as HARs that were initially stable but then changed a million years ago. While her lab has now been able to confirm that it was tweaks in the HAR that shaped our brain, the team is yet to understand the role they play in making humans distinct from other primates.

For enhancers in particular, the way the genome folds in three dimensions, much like origami, is crucial. This is due to the fact that enhancers can affect any nearby gene’s activity, which varies based on how DNA is folded, says Pollard, a professor at UC San Francisco.

The scientists employed machine learning, a type of artificial intelligence, on human and chimpanzee DNA sequences in order to predict DNA folding patterns. This made it easier for scientists to pinpoint the parts of the human genome that fold differently from chimpanzees, and the AI estimated that about 30% of HARs were located in these regions.

The researchers added that there might have been grave repercussions for our ancestors if human DNA close to HARs folded differently and brought different genes close to HARs.
